Selective targeting of striatal parvalbumin-expressing interneurons for transgene delivery

چکیده
PVCre mice--> combined with AAV-FLEX vectors allowed efficient and specific targeting of PV+ interneurons in the striatum. However, diffusion viral particles to globus pallidus caused massive transduction projection neurons subsequent anterograde transport transgene product subthalamic nucleus substantia nigra pars reticulata. Different AAV serotypes (1 9) promoters (CBA human synapsin) were evaluated. The combination AAV1, a moderate expression level (human synapsin promoter) precise adjustment stereotaxic coordinates anterior dorsolateral part striatum necessary avoid GP neurons. Even absence direct due particles, could be retrogradely transduced via their terminals present dorsal diffusion, GP-Str poorly or not suggesting that retrograde did significantly impair selective striatal Finally, prominent reduction number (about 50 %) was evidenced presence Cre recombinase functional effects AAV-mediated mice should analyzed caution.

EGF amplifies the replacement of parvalbumin-expressing striatal interneurons after ischemia.
EGF promotes proliferation and migration of stem/progenitor cells in the normal adult brain. The effect of epidermal growth factor on neurogenesis in ischemic brain is unknown, however. Here we show that intraventricular administration of EGF and albumin augments 100-fold neuronal replacement in the injured adult mouse striatum after cerebral ischemia. Newly born immature neurons migrate into t...
متن کاملGlobus Pallidus Externus Neurons Expressing parvalbumin Interconnect the Subthalamic Nucleus and Striatal Interneurons
The globus pallidus externus (GP) is a nucleus of the basal ganglia (BG), containing GABAergic projection neurons that arborize widely throughout the BG, thalamus and cortex. Ongoing work seeks to map axonal projection patterns from GP cell types, as defined by their electrophysiological and molecular properties. Here we use transgenic mice and recombinant viruses to characterize parvalbumin ex...
متن کاملParvalbumin-Expressing Interneurons Linearly Transform Cortical Responses to Visual Stimuli
The response of cortical neurons to a sensory stimulus is shaped by the network in which they are embedded. Here we establish a role of parvalbumin (PV)-expressing cells, a large class of inhibitory neurons that target the soma and perisomatic compartments of pyramidal cells, in controlling cortical responses. By bidirectionally manipulating PV cell activity in visual cortex we show that these ...
متن کاملParvalbumin-Expressing Interneurons Linearly Control Olfactory Bulb Output
In the olfactory bulb, odor representations by principal mitral cells are modulated by local inhibitory circuits. While dendrodendritic synapses between mitral and granule cells are typically thought to be a major source of this modulation, the contributions of other inhibitory neurons remain unclear. Here we demonstrate the functional properties of olfactory bulb parvalbumin-expressing interne...
متن کاملExcitatory and inhibitory synapses in neuropeptide Y-expressing striatal interneurons.
Although rare, interneurons are pivotal in governing striatal output by extensive axonal arborizations synapsing on medium spiny neurons. Using a genetically modified mouse strain in which a green fluorescent protein (GFP) is driven to be expressed under control of the neuropeptide Y (NPY) promoter, we identified NPY interneurons and compared them with striatal principal neurons. We found that ...
